Tucker had a golden chance to open his account in only the second minute when Devon were awarded a penalty for a foul on Mike Smith, but his spot kick was saved by the Gloucestershire goalkeeper.
It did not take long for the former Barnstaple Town player to make amends.
On 17 minutes, he delivered a free kick that deceived the visiting defence and floated into the net.
Tucker drilled in the second from 20 yards on 76 minutes.
Gloucestershire finished the game with nine men, having suffered injuries after using all their substitutes.
And Tucker, who has scored eight goals for Ilfracombe this season, sealed Devon's win in the eighth minute of stoppage time.
He was brought down in the area and picked himself up to convert the penalty.
Berkshire and Buckinghamshire beat Dorset 4-1 to reach the final of the South and West counties knockout competition.
Liam Gregory and Lewis Goss, of Barnstaple Town, were named in the Devon under-18 squad who came back from 2-0 and 3-1 to draw 3-3 with Somerset on Tuesday.
Mollie Jones, who plays for Barnstaple FC and Exeter College Academy, made a promising debut for the Devon under-18 women's team in their 2-0 home defeat to Somerset in the Bluefin South and West Counties Championship last Thursday.
