Two men, aged in their 20s, have been arrested in connection with the collision.
Devon and Cornwall Police confirmed at 1.30pm (Friday) that both men are being investigated for causing death by dangerous driving. They are currently in police custody at Barnstaple Police Station.
One man had initially been arrested for dangerous driving, while a second man had been arrested on suspicion of aiding and abetting a disqualified driver.
The crash, involving a black Rover 25 which contained a number of people, occurred on a B-road at Loxhore Cross, north east of Barnstaple.
It is understood that the vehicle veered of the road and rolled 25ft down a steep embankment before catching on fire.
A spokesperson for Devon and Cornwall Police said: "A small community has lost two young people and our thoughts are with their families and friends at this tragic time.
“Collision investigations are taking place at the scene and we anticipate the road will remain closed for some hours. This is the early stages of an investigation and no conclusions should be drawn about the incident at this time.”
At present the road is closed between Bratton Cross and Snapper with local diversions in place.
It is likely that the road will be shut for some time.
More details will appear as we have them.

Another busy road also remains closed in Barnstaple following a separate crash this morning.
The A39 is closed between Roundswell and Lake Roundabout after a crash involving one vehicle.
